RESUMO

The application of microbial fertilizer plays an important role in improving soil restoration and fertilizer utilization. The effects of microbial fertilizer are greatly affected by crop genotypes and ecological conditions. Little is known about the effects of microbial fertilizers on maize production in Northeast China. To develop microbial fertilizer specific to the black soil and the climate characteristics of Northeast China, we isolated five plant rhizosphere-promoting bacteria (PGPR), named as MZ1, MZ2, MZ3, MZ4 and MZ5, with different degrees of biological functions such as IAA synthesis, phosphate-solubilizing, potassium-solubilizing and siderophore-releasing, from the rhizosphere of maize field. The analysis of ecological adaptability showed that those five strains differed in salt resistance, drought tolerance, acid and alkali resistance, pesticide resistance. The 16S rRNA gene sequences analysis showed that the strains MZ1, MZ2, MZ3, MZ4 and MZ5 belonged to the genus of Sphingomonas, Enterobacter, Pseudomonas, Bacillus and Rhizobium, respectively. In maize field experiment with 50% nitrogen fertilizer reduction, the inoculation with MZ1, MZ3 and MZ5 increased grain yield by 19.9%-25.0%. MZ1, MZ3, and MZ5 could be used as microbial fertilizers for maize in Northeast China.

Glucokinase (GK) activity, which is rapidly regulated by glucokinase regulatory protein (GKRP) in the liver, is crucial for blood glucose homeostasis. In this paper, the GK activation mechanisms of 1-deoxynojrimycin (DNJ), resveratrol (RES), oxyresveratrol (OXY), cyanidin-3-glucoside (C3G), and cyanidin-3-rutinoside (C3R) were compared. The results revealed that DNJ, RES, C3G, and C3R could differently improve glucose consumption and enhance intracellular GK activities. DNJ and RES significantly promoted GK translocation at 12.5 µM, whereas other ingredients showed moderate effects. DNJ, C3G, and C3R could rupture intramolecular hydrogen bonds of GK to accelerate its allosteric activation at early stage. RES and OXY could bind to a "hydrophobic pocket" on GK to stabilize the active GK at the final stage. Otherwise, RES, OXY, C3G, and C3R could interact with GKRP at the F1P binding site to promote GK dissociation and translocation. Enzymatic assay showed that RES (15-50 µM) and OXY (25-50 µM) could significantly enhance GK activities, which was caused by their binding properties with GK. Moreover, the most dramatic up-regulation effects on GK expression were observed in C3G and C3R groups. This work expounded the differences between GK activation mechanisms, and the new findings would help to develop new GK activators.

Bamboo leaf extract as a food additive has been used for preventing the oxidation of food. In the present study, we investigated the influence of Phyllostachys edulis leaf extract on starch digestion. Orientin, isoorientin, vitexin, and isovitexin were determined as its α-amylase inhibitory constituents. An inhibitory kinetics experiment demonstrated that they competitively inhibit α-amylase with Ki values of respectively 152.6, 11.5, 569.6, and 75.8 µg/mL. Molecular docking showed the four flavones can interact with the active site of α-amylase, and their inhibitory activity was greatly influenced by the glucoside linking position and 3'-hydroxyl. Moreover, the results of starch-iodine complex spectroscopy, X-ray diffraction, and scanning electron microscopy indicated that P. edulis flavonoids retard the digestion of starch not only through interaction with digestive enzymes, but also through interaction with starch. Thus, P. edulis leaf extract can be potentially used as a starch-based food additive for adjusting postprandial hyperglycemia.

OBJECTIVE: To study the optimal waist circumference (WC) cut-off values for identifying metabolic risk factors in middle-aged and elderly subjects in Shandong Province of China. METHODS: A total of 2,873 men and 5,559 women were included in this cross-sectional study. Metabolic syndrome (MetS) was diagnosed according to the definition of Chinese Diabetes Society in 2004. The relation between WC and MetS was analyzed by multivariate logistic regression analysis. The optimal WC cut-off values were identified using the area under the ROC curve and the different diagnostic criteria for central obesity were compared. RESULTS: The WC was the risk factor for MetS independent of BMI, blood glucose, blood lipid, and blood pressure. The optimal WC cut-off value was 83.8 cm and 91.1 cm for identifying MetS in women and men, respectively. Compared with 80 cm and 85 cm for women and men, 85 cm and 90 cm had a higher Youden index for identifying all metabolic risk factors and MetS in women and men. CONCLUSION: The appropriate WC cut-off value is 85 cm and 90 cm for identifying central obesity and MetS in women and men in Shandong Province of China.

DNA methylation is an important and reversible epigenetic modification which regulates genomic stability. Methylation is essential for mammalian development. Generally, gene expression level and DNA methylation are negative correlation. Transcriptional silencing via methylation of CpG islands in the promoter is important for cell growth and differentiation and plays a key role in tumorigenesis. Demethylation drug can modify chromatin and restore the ability of anti-oncogene. Demethylation therapy as a new therapy may treat efficiently hematological malignancies with resistance and relapse. In this review, DNA methylation mechanism, relationship between aberrant methylation and hematologic malignancies, mechanism of demethylation therapy, the advance of research on the demethylation therapy of hematological malignancies, such as acute and chronic leukemia, lymphoma, myelodysplastic syndrome were summarized.
